چکیده

Rice waxy gene codes for the starch granule-bound starch synthase (EC 2.4.1.11) (1). To characterize its molecular structure, a rice {Oryza sativa subsp. japonica Hengfeng cultivar) genomic library was constructed in XEMBL-3 vector. Two overlapping genomic clones containing waxy gene sequence were screened and identified from this library by using maize waxy gene DNA as probe (kindly provided by Dr N.Fedoroff). The nucleotide sequences of rice waxy gene and its 5' and 3' flanking regions (5499 bp) are presented here. Alignment of nucleotide sequence of waxy gene from rice with that of maize (2) and barley (3) revealed the presence of 13 introns and 14 exons. The fulllength of waxy preprotein is 609 amino acid residues. ACKNOWLEDGEMENTS

Genotyping common SNP and a microsatellite sequence closely linked to waxy gene in rice by DNA based markers

The potential of different DNA based molecular markers was examined for the detection of single nucleotide polymorphism (SNP) in the waxy gene and a microsatellite (SSR) sequence closely linked to it in a collection of rice varieties. DNA was extracted from leaf samples of 68 different rice cultivars by the CTAB method and specific primers were designed for the amplification of waxy gene and SS...

Waxy gene haplotypes: Associations with apparent amylose content and the effect by the environment in an international rice germplasm collection

Apparent amylose content (AAC), the key determinant of rice end-use quality attributes, is primarily controlled by the Waxy gene which codes for granule bound starch synthase (GBSS). We examined the combination of sequence variation in the Waxy gene and environmental effects, and their associations with AAC using 171 rice accessions originating from 43 countries. The combination of two single-n...

Nonsense-mediated decay of mutant waxy mRNA in rice.

Two rice (Oryza sativa) waxy mutations of the Japonica background were shown to contain approximately 20% of the fully spliced mRNA relative to the wild type. Sequencing analysis of the entire waxy genes of the two mutants revealed the presence of premature translation termination codons in exon 2 and exon 7. These results indicated that the lower accumulation of fully spliced RNA in the mutant...

Correlation analysis between starch properties and single nucleotide polymorphisms of waxy genes in common rye (Secale cereale L.).

To understand the relationships between single nucleotide polymorphisms (SNPs) in the waxy gene and starch parameters in common rye, we performed sequence characterization, enzyme activity testing, amylopectin/amylose ratio evaluation, starch property testing, and correlation analysis. Specific primers were used to clone waxy from 20 rye cultivars. Sequence analysis showed that waxy was 2852 bp...

Selection under domestication: evidence for a sweep in the rice waxy genomic region.

Rice (Oryza sativa) was cultivated by Asian Neolithic farmers >11,000 years ago, and different cultures have selected for divergent starch qualities in the rice grain during and after the domestication process. An intron 1 splice donor site mutation of the Waxy gene is responsible for the absence of amylose in glutinous rice varieties. This mutation appears to have also played an important role...
